Studies have shown that there is an inverse correla¬tion between smooth muscle proliferation and cyclin-dependent kinase inhibitors (CKIs) such as P21 and P27. Gene transfer methods that result in overexpression of CKIs reduce smooth muscle cell proliferation and neointimal formation. P1 and P27 knock-out mice given a Western diet undergo accelerated arterial cell prolifera¬tion and atherosclerosis.
B. Other Effects
Significant clinical trial-related deaths have occurred. The potential for vector-induced cytotoxicity remains. Prolif-erative retinopathy and retinal hemorrhage occur and caution is warranted.
